GeoMx® Digital Spatial Profiler (DSP)

Morphologically guided proteomic and transcriptomic Spatial Profiling.

Nanostring logo

The GeoMx DSP allows researchers to spatially profile complex microenvironments by using fluorescent staining patterns as a mask to profile expression in certain tissue types or cell populations.

  • Standard IHC slides e.g. SuperFrost Plus
  • 5 µM (FFPE) or 10 µM (Fresh/Fixed Frozen)
  • Stain up to 4 morphology markers (FITC/Cy3/Texas Red/Cy5) to reveal the structural architecture and identify regions of interest (ROI)
  • UV cleavable oligonucleotide tags are released from user-defined Areas of Illumination (AOI) via UV exposure and are collected into 96 well plates, each well relating to a single AOI.
  • For larger panels the 27bp oligonucleotide tags are sequenced: Whole Transcriptome Atlas (18,000 / 22,000 genes), Cancer Transcriptome Atlas (1,800 / 1,962 genes), IO Proteome (570 proteins), Neural Proteome Atlas (650 proteins) & Discovery Proteome Atlas (1200 proteins)
  • For small panels the photocleaved oligos are hybridised to a reporter probe and imaged on the nCounter: Immune pathways (96 genes), Immuno-oncology (up to 96 proteins), Neuroscience (up to 96 proteins)

Example of possible segmentation strategies
Example of possible segmentation strategies:
A: SOX 2 positive cells only (1 AOI per ROI)
B: comparison of tumour with immediate TME (2 AOI per ROI)
